LRRC45 also known as Leucine-Rich Repeat-Containing Protein 45 is a component of the centrosome an organelle critical for cell division. The protein has a molecular mass of about 91 kDa. It is ubiquitously expressed throughout various tissues and cells especially those with a rapid turnover. The main mechanical role of LRRC45 is organizing microtubules during the cell cycle ensuring correct chromosome segregation. Its alternate configuration sometimes noted as C10orf90 may have similar functional characteristics.
Biological function summary
LRRC45 is an important component in maintaining centrosome integrity. It functions as part of the centrosomal protein complex where it interacts closely with other centrosomal proteins to regulate their localization and stability. By stabilizing microtubules LRRC45 ensures proper anchoring of spindle fibers and helps mediate centrosome cohesion. These actions are essential for accurate mitosis and cellular proliferation.
Pathways
LRRC45 plays a role in the cell cycle and specifically in the centrosome cycle. It participates in pathways involving centrosome duplication and separation integral for proper cell division. It works closely with proteins such as Cep152 and Plk4 which initiate centrosome biogenesis. The integration of LRRC45 within these pathways highlights its importance in maintaining genomic stability across cell generations.
Defects or mutations in LRRC45 have been linked to certain forms of cancer and microcephaly. The protein's vital role in centrosome dynamics positions it as a point of interest in disorders characterized by aberrant cell division. It interacts with proteins including CEP63 that are implicated in similar pathologies suggesting a complex network where LRRC45 plays a significant role. Understanding its contributions to these disorders may provide insights into novel therapeutic approaches.
