SPAG17 also known as sperm-associated antigen 17 is a protein with an approximate mass of 220 kDa. This protein is mainly expressed in testicular tissues and is abundant in sperm cells as well as other ciliated cells in the respiratory tract. Mechanically SPAG17 is a structural component of the axoneme a core part of cilia and flagella where it plays a role in maintaining the integrity of these structures.
Biological function summary
There exist strong associations between the workings of SPAG17 and the motility of sperm and cilia. It is a part of the central apparatus complex of the axoneme which is important for normal ciliary and flagellar beat patterns. In sperm cells SPAG17 impacts sperm motility greatly influencing male fertility. In the respiratory system its role extends to aiding clearance of mucus through ciliary motion thereby helping maintain respiratory function.
Pathways
SPAG17 participates in the pathways regulating ciliary motion and spermatogenesis. The involvement of SPAG17 in these pathways highlights its relationships with other axonemal components such as dynein heavy chains which are responsible for motor activity along microtubules. Additionally it fits into the broader network of proteins governing microtubule dynamics working in concert to achieve its structural and functional roles.
SPAG17 influences conditions related to ciliary dysfunction such as primary ciliary dyskinesia. This disorder arises from defects in the motility of cilia manifesting in respiratory issues and potentially reduced fertility. The protein's relationship with other axonemal proteins like dynein motors links SPAG17 mutations to these symptoms. Additionally alterations in SPAG17 have been explored in research related to male infertility with studies suggesting that impaired sperm motility due to SPAG17 function can contribute to this condition.
