The ARL4C protein also known as ADP-ribosylation factor-like 4C is a small GTPase member of the ARF family with a molecular mass of approximately 20-25 kDa. This protein plays roles in regulating cytoskeletal organization and intracellular trafficking. ARL4C is expressed in various tissues including the brain kidney and liver where it contributes to cellular processes. Its expression levels can vary depending on the physiological conditions and requirements of specific tissues.
Biological function summary
ARL4C contributes to cellular proliferation and migration. It often associates with plasma membranes and cytosolic complexes participating in the regulation of cytoskeletal dynamics through its interaction with other actin-modulating proteins. Additionally ARL4C modulates the activity of signaling pathways that involve small GTPases exerting influence over cell shape and movement which are important for maintaining cellular structure and facilitating response to external stimuli.
Pathways
ARL4C plays an integrating role in the Wnt and MAPK signaling pathways. In the Wnt signaling pathway ARL4C interacts with proteins such as Dishevelled influencing cell fate determination and embryonic development. In the MAPK pathway it has connections that affect cellular responses to growth factors and stress signals interacting with kinases like ERK1/2 which modulate cell division and survival. The involvement of ARL4C in these pathways highlights its importance in regulating diverse biological responses.
ARL4C has been linked to cancer and fibrosis. Overexpression of ARL4C has been observed in several cancers where it promotes tumor cell migration and invasion suggesting a potential role in cancer progression. In pulmonary fibrosis altered ARL4C expression affects the fibrotic process influencing tissue repair mechanisms. In both cases interactions with proteins such as Rac1 and other family GTPases are critical as they modulate various cellular processes related to disease development and progression.
