TYSND1 also known as Trypsin-5 is a serine protease enzyme with a molecular mass of approximately 46 kDa. It plays an important role in catalyzing the cleavage of peptide bonds in various substrate proteins. TYSND1 primarily localizes in the peroxisomes and mitochondria where it is highly expressed. In particular it is found in tissues with high metabolic activities like the liver and kidney suggesting it has functions in energy metabolism and cellular detoxification processes.
Biological function summary
TYSND1 contributes to the maintenance of protein homeostasis by participating in the processing of proteins that are imported into peroxisomes. It forms part of a larger complex involved in the maturation and processing of peroxisomal matrix proteins. This process ensures that imported proteins achieve their proper functional forms which is important for peroxisome function. Additionally TYSND1 aids in the cleavage of the enzyme PTS1 which signals protein import into peroxisomes therefore influencing peroxisomal protein composition and activity.
Pathways
TYSND1 plays important roles in the peroxisomal protein import pathway and the fatty acid β-oxidation pathway. In the import pathway TYSND1 works with other proteins such as PEX11 and PEX13 to facilitate the correct targeting and processing of enzymes that execute peroxisomal functions. In the fatty acid β-oxidation pathway TYSND1 acts indirectly by ensuring the availability and proper configuration of enzymes that catalyze the breakdown of fatty acids which is an important energy production process.
Dysregulation of TYSND1 activity has links to disorders like peroxisome biogenesis disorders and fatty liver disease. Peroxisome biogenesis disorders result from impaired import of necessary enzymes into peroxisomes potentially linked to dysfunctional TYSND1 activity. Additionally alterations in TYSND1 expression are associated with metabolic conditions such as fatty liver disease where it interacts with proteins like PPARα that regulate lipid metabolism.
