UBXN2B also known as UBX domain-containing protein 2B is a human protein with a molecular mass of approximately 60 kDa. It contains a UBX domain that enables interactions with various cellular components. UBXN2B is largely expressed in the cytoplasm and nucleus of cells. The protein acts as an adaptor in the ubiquitin-proteasome system facilitating the delivery of ubiquitinated substrates for degradation. It also interacts with p97/VCP an important AAA ATPase involved in extracting misfolded proteins.
Biological function summary
UBXN2B plays a significant role in the endoplasmic reticulum-associated degradation (ERAD) pathway managing protein quality control. It is an essential part of protein complexes that regulate ER-associated protein degradation. These complexes ensure proper folding and elimination of misfolded proteins which helps maintain cellular homeostasis. Lack of proper function of such complexes can lead to accumulation of defective proteins impacting cell health.
Pathways
UBXN2B participates in the protein degradation pathways including the ERAD and ubiquitin-proteasome pathways. It works closely with the p97/VCP complex to recognize and dismantle faulty proteins. UBXN2B also associates with other proteins in the degradation system such as ubiquitin ligases which tag unwanted proteins for destruction. This facilitates the cycling and reuse of cellular components essential for cellular metabolism and function.
UBXN2B has been linked to neurodegenerative diseases and cancer. Misregulation may contribute to conditions like Parkinson’s disease where protein aggregation negatively affects neurons. In certain cancers altered UBXN2B expression influences tumor growth by disrupting normal protein degradation pathways. These conditions often involve proteins like p97/VCP which under normal circumstances works with UBXN2B to maintain cellular proteostasis.
