Treslin also known as TICRR is a protein that plays an important role in initiating DNA replication. It has a molecular mass of approximately 220 kDa. Treslin is expressed in various human tissues with significant levels observed in cell types that actively undergo division. Mechanically Treslin primarily functions in the S phase of the cell cycle enabling the transition from the G1 phase to the S phase. It does so by interacting with other replication factors facilitating the assembly of the pre-replicative complex necessary to commence DNA synthesis.
Biological function summary
Treslin holds a significant position in the progression of the cell cycle. It participates in complex formation with other proteins such as TopBP1 where it supports replication initiation by recruiting and stabilizing additional replication machinery components. Treslin is necessary for the loading of Cdc45 onto replication origins. It ensures precise control over DNA replication preventing unscheduled or erroneous replication events which can lead to genomic instability.
Pathways
Treslin operates in pivotal DNA replication and cell cycle control pathways. It works closely with proteins like Cdc45 and TopBP1 within the DNA replication start pathway and the cell cycle's S-phase entry process. Treslin's interactions with these proteins ensure that DNA replication starts accurately within the cell cycle framework thereby maintaining genomic integrity and proper cell function.
Researchers associate Treslin with cancer development and progression. Its malfunction or aberrant expression can result in genomic instability promoting oncogenesis. Treslin's interaction with TopBP1 also ties it to disorders involving cellular cycle dysregulation. Understanding Treslin's role in these disease pathways may help design therapeutic strategies that target replication machinery components to inhibit cancer cell proliferation.
