Engrailed and Invected proteins often studied using the alternate name Engrailing function as homeodomain transcription factors. They play key roles in regulating gene expression and pattern formation during embryonic development. The Engrailed proteins have a molecular mass around 37 kDa and are expressed in several tissues including the central nervous system of Drosophila melanogaster. These proteins share structural similarities and often operate cooperatively in their functions.
Biological function summary
These transcription factors govern the segmentation and patterning processes critical for proper development. Engrailed and Invected proteins form part of the Engrailed/Hedgehog signaling complex which is important for maintaining the anterior-posterior axis in developing embryos. Through their interactions with other transcription factors and signaling molecules they ensure proper spatial and temporal expression of target genes important for tissue differentiation.
Pathways
A significant role for Engrailed and Invected proteins lies in the Hedgehog and Wingless signaling pathways. Engrailed assists in activating the Hedgehog pathway affecting cell fate decisions and patterning. Additionally Engrailed proteins interact with the Wingless signaling pathway where coordination with proteins like Cubitus interruptus leads to precise genetic regulation vital for maintaining developmental boundaries.
Abnormal expression or mutations in these proteins link to various developmental disorders and diseases. Misregulation of Engrailed and Invected can contribute to neoplastic conditions and disorders of the central nervous system. For instance links with proteins such as Patched and Smoothened in the Hedgehog pathway reveal their involvement in certain basal cell carcinomas when pathways fail to regulate cell growth properly.
