NKIRAS2 also known as Nuclear factor kappa B Inhibitor-interacting Ras-like protein 2 is a protein with estimated molecular mass around 21 kDa. This protein acts as a negative regulator of NF-kB signaling pathways by preventing the phosphorylation and degradation of IkB proteins. It serves as a molecular switch in the inhibition of Ras-related proteins and shows expression in various tissues notably in immune cells and the brain.
Biological function summary
NKIRAS2 functions to modulate inflammatory responses and immune regulation. It is observed interacting within a larger complex that includes NF-kB and IkB proteins. By binding to these molecules NKIRAS2 plays a role in controlling the activation of transcription factors important for immune and stress response pathways. This modulation prevents excessive immune responses and maintains cellular homeostasis.
Pathways
NKIRAS2 involvement is most noted in the NF-kB signaling pathway which is essential for immune response regulation and the Ras signaling pathway contributing to the regulation of cell proliferation and survival. NKIRAS2 acts in coordination with proteins like IkB and Ras family members to modulate the activity of these signaling pathways. This interaction helps finely tune cellular responses to external stimuli protecting cells from unwarranted activation.
NKIRAS2 has links to inflammatory diseases and certain types of cancer. Its role in the inhibition of NF-kB pathways makes it a point of interest in pathologies where this signaling becomes dysregulated. Deregulation of NKIRAS2 or its interacting partners such as IkB proteins can lead to chronic inflammation and has associations with cancers where NF-kB signaling is aberrantly activated. Research into these pathways could potentially lead to novel therapeutic strategies.
