PI3K (p110 delta/p85 alpha) also known as phosphoinositide 3-kinase delta and alpha is composed of two subunits: the catalytic p110 delta and the regulatory p85 alpha. The complex has a molecular mass of about 124 kDa for p110 delta and 83 kDa for p85 alpha. PI3K is mainly expressed in leukocytes where it plays roles in immune responses. Its activity involves phosphorylation of phosphoinositides at the 3-position of the inositol ring acting as an important player in signal transduction.
Biological function summary
PI3K influences cell growth survival and metabolism by producing lipid second messengers. It partners with other PI3K proteins including p85 PI3K forming heterodimeric complexes. PI3K delta is important in hematopoietic cells promoting cellular responses to stimuli and modulating immune system activity. Dysfunction in these activities can lead to immune system disorders. The dual role of PI3K in both regulating normal cellular processes and contributing to pathologies makes it an attractive therapeutic target.
Pathways
PI3K is a central component of the PI3K/AKT signaling pathway which is significant in mediating cellular proliferation and survival. This pathway interacts with other signaling molecules such as the mTOR pathway to manage cellular growth and metabolism. PI3K works closely with proteins like PTEN a phosphatase that negatively regulates the pathway and AKT a downstream effector. Understanding these interactions highlights their importance in maintaining cellular homeostasis and exploring therapeutic strategies.
Dysregulation of PI3K activity links to cancer and inflammatory diseases. In cancer mutations or overexpression of PI3K delta can lead to uncontrolled cell proliferation. Targeting PI3K with inhibitors offers potential for cancer treatment. In inflammatory disorders altered PI3K signaling can result in abnormal immune responses suggesting its role in diseases like rheumatoid arthritis. Researchers focus on modulating PI3K activity to develop treatments for these conditions particularly addressing its interactions with proteins like mTOR and PTEN.
