PAXX also known as C9orf142 is a component of the non-homologous end joining (NHEJ) pathway which is essential for DNA double-strand break repair. The protein has a molecular mass of approximately 25 kDa. Researchers have found that PAXX is widely expressed in various tissues indicating its physiological importance across different cell types.
Biological function summary
PAXX plays an important role as a component of the XLF Ku70 and DNA-PKcs complex during the NHEJ process. This process is a primary mechanism to repair double-strand breaks in DNA maintaining genomic stability. PAXX achieves this by facilitating the joining of broken DNA ends. This function ensures the accuracy and efficiency of the repair process which is an important aspect of maintaining cellular health and preventing mutations.
Pathways
PAXX participates in the DNA repair pathways specifically the NHEJ pathway. It works closely with other proteins like Ku70 and DNA-PKcs which recognize and bind to the DNA ends to initiate the repair process. PAXX interacts with these proteins to stabilize the repair complex therefore ensuring completion and fidelity of DNA repair and subsequently it safeguards the genome against instability.
Defects or malfunctions in PAXX can lead to a predisposition to cancer due to weakened DNA repair capabilities. Its involvement in NHEJ makes it critical for preventing genomic instability a hallmark of cancerous cells. Additionally PAXX has connections to XLF and DNA-PKcs where mutations or alterations in these proteins can exacerbate similar disorders highlighting the interconnected network of DNA repair proteins in maintaining cell integrity.
