DEFB119 also known as beta-defensin 119 is a small antimicrobial peptide with a mass of approximately 9 kDa. This peptide belongs to a larger family of beta-defensins which are structurally similar in forming a beta-sheet due to conserved cysteine residues. DEFB119 expression occurs predominantly in epithelial tissues including those of the respiratory and urogenital tracts. It serves an essential role by imparting innate immune defense through its antimicrobial properties against various pathogens including bacteria and fungi.
Biological function summary
DEFB119 contributes to the host defense by disrupting pathogen cell membranes through electrostatic interactions leading to cell lysis. It does not operate as part of a large complex but functions independently within the innate immune responses. By interacting directly with microbial membranes DEFB119 effectively neutralizes these pathogens reducing infection and maintaining tissue homeostasis. Additionally it may influence inflammatory responses and wound healing further contributing to its protective functions in epithelial surfaces.
Pathways
The DEFB119 protein integrates into the innate immune response pathways specifically those related to antimicrobial defense. It plays a role within the Toll-like receptor signaling pathway where it modulates immune responses against microbial invasion. DEFB119 is functionally related to other antimicrobial peptides and proteins such as DEFB4 and LL-37 which collaborate in the antimicrobial activity and immune modulation by acting in similar pathways. The signaling triggered by these pathways often results in protective responses essential for maintaining epithelial integrity.
DEFB119 has shown connections to conditions involving immune system dysfunctions such as cystic fibrosis where epithelial barrier defenses are compromised. In such disorders altered expression of DEFB119 may contribute to the increased risk and persistence of infections commonly seen in patients. Additionally aberrations in DEFB119 expression or function can relate to inflammatory bowel disease where epithelial inflammation and immune responses are dysregulated. This places DEFB119 in a network with other defensins and immune-regulating proteins that are involved in maintaining balanced immune reactivity and intestinal health.
