ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of Human KLK in cell culture supernatants, serum, plasma (heparin , EDTA), human milk and saliva.
A KLK5 specific mouse monoclonal antibody has been precoated onto 96-well plates. Standards and test samples are added to the wells and incubated. A biotinylated detection polyclonal antibody from goat specific for KLK5 is then added followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. TMB is then used to visualize the HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the Human KLK5 amount of sample captured in plate.

KLK5 also known as Kallikrein-related peptidase 5 is a serine protease with an estimated molecular mass of approximately 28 kDa. It plays a role in skin homeostasis. The protein is predominantly present in tissues like the skin mammary gland and prostate gland. KLK5 is secreted as an inactive zymogen and requires proteolytic activation. Its primary site of action is the extracellular space where it participates in cleaving peptide bonds after lysine and arginine residues.
Biological function summary
The function of KLK5 involves maintaining the skin's barrier function and facilitating desquamation which is the shedding of the outermost surface of the skin. KLK5 does not typically function as part of a larger protein complex but may interact with and activate other kallikreins such as KLK7 and KLK14. It is implicated in processes like keratinocyte differentiation and inflammation through its proteolytic activities.

KLK5 is integral to skin desquamation and inflammation-related pathways. KLK5 exhibits associations with conditions like Netherton syndrome and atopic dermatitis. Netherton syndrome results from mutations affecting related inhibitors such as LEKTI leading to the dysregulation of KLK5 activity. This excessive proteolytic activity disrupts the skin barrier and contributes to the characteristic scaling and inflammation. Additionally KLK5's influence on inflammation via pathways connected to PAR-2 links it to atopic dermatitis which is characterized by chronic inflammation and itchiness.
