ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of Human soluble TLR2 in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA).
A TLR2 (CD282) specific mouse monoclonal antibody has been precoated onto 96-well plates. Standards and test samples are added to the wells and incubated. A biotinylated detection polyclonal antibody from goat, specific for TLR2 is then added followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. TMB is then used to visualize the HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the Human TLR2 amount of sample captured in plate.

TLR2 also known as Toll-like receptor 2 is a type 1 transmembrane protein playing a role in the innate immune system. This protein has a molecular mass of approximately 90 kDa. It is present on the surface of immune cells such as macrophages dendritic cells and some types of epithelial cells. TLR2 recognizes and binds to pathogen-associated molecular patterns acting as a receptor for lipoproteins and lipopeptides from Gram-positive bacteria mycobacteria and fungi.
Biological function summary
TLR2 works as part of a complex with other proteins like TLR1 or TLR6 to initiate the immune response. It helps detect microbial components which leads to the activation of downstream signaling pathways and subsequent release of pro-inflammatory cytokines.
