CalDAG-GEFI also known as RasGRP2 is a guanine nucleotide exchange factor that plays a critical role in cellular signaling by activating small GTPases like Rap1. The protein consists of 546 amino acids giving it a molecular mass of 61 kDa. It is widely expressed in platelets leukocytes and the brain. The signaling pathways that involve CalDAG-GEFI are essential for the regulation of calcium (Ca2+) levels within cells impacting many cellular functions.
Biological function summary
CalDAG-GEFI influences cell adhesion and migration by activating Rap1 a member of the Ras family of GTPases. This protein does not work alone and often forms part of signaling complexes that help relay external signals to intracellular responses. Through its regulatory effects on Rap1 CalDAG-GEFI affects platelet aggregation and leukocyte trafficking which are important for normal immune function and hemostasis.
Pathways
CalDAG-GEFI plays significant roles in the Ras and Rap1 signaling pathways. It operates with other proteins like PLC (phospholipase C) to mediate calcium-dependent activation of Rap1. This activity links CalDAG-GEFI to the regulation of integrins affecting cellular interactions with the extracellular matrix. This linkage is critical for various cellular activities including immune surveillance and tissue repair processes.
CalDAG-GEFI relates mainly to bleeding disorders and immune system dysfunctions. Its role in platelet aggregation makes it significant in conditions like Glanzmann thrombasthenia where platelet plugs fail to form properly. Furthermore improper regulation of CalDAG-GEFI can contribute to autoimmune diseases due to dysregulated leukocyte migration. This protein's activity is closely associated with integrins which also play roles in these conditions underlining its importance in maintaining normal hemostatic balance and immune response.
