competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of RBP in plasma and serum.
A Retinol Binding Protein specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently biotinylated Retinol Binding Protein is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Complex is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is inversely proportional to the amount of Retinol Binding Protein captured in plate.
The immunogen for the RBP-capture antibody is total RBP and thus the capture antibody should detect both RBP1 and RPB4, among others.
The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

Retinol Binding Protein (RBP) also known as RBP4 is a transport protein with a molecular mass of approximately 21 kDa. It binds retinol (vitamin A) and transports it from the liver storage sites to peripheral tissues via the bloodstream. RBP exhibits expression mainly in the liver although it is also found in other tissues such as the kidneys and adipose tissues. The primary function involves facilitating vitamin A circulation in the body ensuring that retinoid-dependent processes can operate effectively.
Biological function summary
The RBP4 plays an essential role in the regulation of vitamin A homeostasis. It forms a complex with transthyretin which prevents the loss of RBP-retinol in the kidney by inhibiting glomerular filtration. This interaction stabilizes the entire structure allowing efficient retinol delivery to target cells. The tight regulation and functioning of this complex are important for numerous bodily processes that require vitamin A including vision cellular growth and differentiation.
Pathways
The function of RBP4 is significantly associated with the retinol metabolism pathway and the visual cycle pathway. In retinol metabolism it works in tandem with other proteins such as lecithin retinol acyltransferase (LRAT) to control retinol levels in the body. In the visual cycle pathway RBP4 interacts indirectly with visual cycle proteins like opsins to support retinal release from the bloodstream to eye tissues essential for the maintenance of healthy vision.
RBP4 has been linked to conditions such as type 2 diabetes and night blindness. Elevated serum levels of RBP4 are associated with insulin resistance in type 2 diabetes where it may affect glucose metabolism. Additionally disruptions in RBP4 function or levels can lead to vitamin A deficiency contributing to night blindness through impaired retinol supply to the retina. When considering these diseases RBP4 is often studied alongside other proteins such as insulin receptor substrates and rhodopsin which play roles in the respective pathways and conditions.
