MRPL34 also known as 39S ribosomal protein L34 mitochondrial is a component of the mitochondrial ribosome large subunit. It has a molecular mass of approximately 15 kDa. MRPL34 plays a role in mitochondrial protein synthesis and is widely expressed in tissues with high energy demands such as the heart skeletal muscles and brain. The mitochondrial ribosomes differ from cytoplasmic ribosomes in protein-to-rRNA ratio and are essential for proper mitochondrial function.
Biological function summary
MRPL34 is part of the large subunit of the mitochondrial ribosome which assembles with the small subunit to translate mitochondrial-encoded proteins essential for the oxidative phosphorylation system. Within the mitochondrial ribosome which is composed of roughly 50 distinct proteins and 2 ribosomal RNA molecules MRPL34 contributes to the formation and stabilization of the ribosomal RNA core structure. Its involvement in mitochondrial protein translation directly affects cellular energy metabolism and overall mitochondrial function.
Pathways
MRPL34 contributes to the mitochondrial translation pathway an essential part of maintaining the electron transport chain by facilitating the synthesis of core components required for oxidative phosphorylation. Key proteins involved in these processes include cytochrome c oxidase and ATP synthase essential for energy production. MRPL34 ensures the proper assembly of these complexes by aiding the accurate translation of mitochondrial genome-encoded proteins.
Mutations or malfunction of MRPL34 can lead to diseases such as mitochondrial myopathy and neurodegenerative disorders like Parkinson's disease. Abnormal function of MRPL34 disrupts mitochondrial protein production affecting cellular energy levels and function. It can also influence the activity of proteins such as cytochrome c further impacting oxidative stress response and cell survival in tissues with high metabolic activity.
