PRODH2 also known as proline dehydrogenase is an enzyme with a molecular mass of approximately 65 kDa. It functions as a catalyst in the oxidation of proline into pyrroline-5-carboxylate. This enzyme facilitates the first step in the proline degradation pathway. It is a mitochondrial protein widely expressed in various tissues with higher levels detected in the brain liver and kidney.
Biological function summary
PRODH2 is essential for amino acid metabolism particularly in proline catabolism. This enzyme helps regulate the balance of proline levels within cells which is important for cellular homeostasis. Although PRODH2 operates largely as an individual unit it may interact with other enzymes within mitochondrial pathways. This interaction supports its role in producing necessary cellular energy and intermediates.
Pathways
PRODH2 plays an important role in mitochondrial respiration and the interconnected network of amino acid metabolism. It is closely linked with the proline biosynthesis and degradation pathway. This pathway also involves the enzyme pyrroline-5-carboxylate reductase which works in tandem with PRODH2 to maintain amino acid turnover and energy production in the mitochondria.
Alterations in PRODH2 function have been associated with schizophrenia and hyperprolinemia. The former is a neuropsychiatric disorder where PRODH2’s altered activity may disrupt normal brain metabolism. In hyperprolinemia a deficiency or malfunction in PRODH2 leads to an accumulation of proline causing various metabolic complications. Schizophrenia also involves interactions with other proteins like DISC1 which influence neural pathways and can link to metabolic processes.
