The SH3 domain-containing kinase-binding protein 1 (SH3KBP1) also known as Cin85 is a multifunctional adapter protein involved in signal transduction pathways. It has a molecular mass of approximately 85 kDa. SH3KBP1 is expressed in a variety of tissues including the brain heart and lungs reflecting its diverse roles in cellular processes. This protein contains multiple SH3 domains that facilitate the binding with other proteins making SH3KBP1 an important modulator of protein-protein interactions.
Biological function summary
SH3KBP1 plays a role in cellular signaling and endocytosis. It forms part of a complex that participates in the regulation of membrane trafficking where it helps in internalization and down-regulation of receptor tyrosine kinases. SH3KBP1 interacts with other signaling molecules allowing it to coordinate the spatial and temporal dynamics of signaling pathways. This protein’s involvement in endocytic pathways highlights its significance in regulating cellular responses to external stimuli.
Pathways
SH3KBP1 is an integral part of the epidermal growth factor receptor (EGFR) signaling and the B-cell receptor (BCR) signaling pathways. In the EGFR pathway it facilitates receptor internalization affecting processes such as proliferation and survival. SH3KBP1 interacts with proteins like c-Cbl and Grb2 which are key players in receptor endocytosis and signal transduction. Through these interactions it modulates signal cascades that influence numerous cellular outcomes.
SH3KBP1 is implicated in cancer and autoimmune diseases. Its role in EGFR signaling connects it to cancer progression as dysregulation of this pathway can lead to uncontrolled cell growth. In autoimmune diseases SH3KBP1's interaction with BCR signaling components including CD19 and Syk can affect immune cell function and lead to pathological immune responses. Understanding the role of SH3KBP1 in these contexts can inform therapeutic strategies targeting its pathway involvement.
