MRPL3 also known as Mitochondrial Ribosomal Protein L3 functions mechanically as a component of the mitochondrial ribosome specifically the large 39S subunit. This protein plays a role in mitochondrial protein synthesis. MRPL3 has a molecular mass of approximately 35 kDa. It is expressed in the mitochondria across various tissues having a pivotal involvement in the synthesis of proteins essential for mitochondrial energy generation.
Biological function summary
MRPL3 is important for the production of mitochondrial proteins by facilitating the translation of mitochondrial mRNA molecules. It forms part of the mitochondrial ribosome complex which is distinct from cytoplasmic ribosomes due to its protein-rich composition and specific ribosomal RNA components. This unique ribosome composition allows MRPL3 to contribute to mitochondrial-specific translational mechanisms that support cellular respiration and energy production.
Pathways
MRPL3 is notably involved in mitochondrial translation and oxidative phosphorylation pathways. In the context of oxidative phosphorylation MRPL3 works alongside other mitochondrial ribosomal proteins like MRPS23 to ensure the proper translation of essential components of the electron transport chain. This pathway is important for ATP production which cells use as an energy source. Efficient functioning of these pathways relies on proteins produced through the process that involves MRPL3.
MRPL3 is associated with mitochondrial diseases and certain forms of cancer due to its role in energy metabolism and protein synthesis. Mutations or dysregulation in MRPL3 may lead to mitochondrial dysfunction resulting in conditions such as mitochondrial cardiomyopathy. In some cancers MRPL3 expression levels can alter linked with changes in cellular energy demands and protein synthesis. The protein MRPL44 has shown to interact with MRPL3 suggesting a contribution to the pathology when mitochondrial protein synthesis is impaired.
