The mGluR1 also known as metabotropic glutamate receptor 1 is a protein that plays an important mechanical role in the central nervous system. It belongs to the G-protein coupled receptor (GPCR) family and serves as a receptor for the neurotransmitter glutamate. The protein has a mass of approximately 132 kDa. It is expressed mainly in the brain particularly in the cerebellum hippocampus and cortex. In these regions mGluR1 is involved in mediating excitatory neurotransmission by modulating ion channels and intracellular signaling pathways.
Biological function summary
MGluR1 contributes to synaptic plasticity a critical process for learning and memory. It forms part of a signaling complex that interacts with several other proteins including Homer proteins which facilitate the link to intracellular calcium release channels. The receptor's activation leads to a cascade of intracellular events including the mobilization of calcium and activation of various kinases. These processes subsequently affect long-term potentiation and depression which are key for adaptable and responsive neural circuits.
Pathways
MGluR1 operates within important signaling pathways such as the phosphoinositide pathway and the MAPK/ERK pathway. In the phosphoinositide pathway it activates phospholipase C which results in the production of inositol trisphosphate and diacylglycerol second messengers that regulate calcium release and protein kinase C activation respectively. Within the MAPK/ERK pathway mGluR1 activation contributes to gene expression changes through ERK activation influencing cellular responses and adaptations. Proteins such as calcium-calmodulin and GRK2 are related to mGluR1 within these pathways impacting receptor sensitivity and function.
MGluR1 plays a role in neurodegenerative disorders like Alzheimer's disease and movement disorders such as spinocerebellar ataxia. In Alzheimer’s disease dysregulation of glutamate signaling involving mGluR1 can contribute to synaptic dysfunction and the progression of neurodegeneration. Spinocerebellar ataxia involves alterations in motor coordination linked to mGluR1 function in the cerebellum. Additionally mGluR1 has associations with proteins such as NMDA receptors in Alzheimer's where altered signaling can exacerbate disease symptoms and progression through excitotoxic pathways.
