RbBP5 also known as Retinoblastoma binding protein 5 acts as a core component of the Set1/COMPASS complex which plays a role in histone methylation. The RbBP5 protein weighs approximately 58 kDa and it is found throughout various tissues with notable expression in areas engaged in active transcription. This widespread presence suggests its substantial impact on chromatin modification processes.
Biological function summary
RbBP5 functions to assist in the regulation of gene transcription. It is an integral part of the Set1/COMPASS complex which is responsible for the tri-methylation of histone H3 at lysine 4 (H3K4me3) a modification associated with transcriptionally active genes. By participating in this complex RbBP5 contributes to the establishment and maintenance of euchromatin regions thereby allowing transcription factors and other proteins involved in gene expression to access DNA more easily.
Pathways
RbBP5 plays a role in chromatin remodeling and transcription regulation pathways. Its involvement in the H3K4me3 modification process places it in the TRX pathway influencing various gene expression patterns. RbBP5 interacts with other components of the Set1/COMPASS complex including ASH2L and WDR5 which collaboratively modulate the levels of histone methylation and ultimately affect gene transcription activities.
RbBP5 has connections to cancer and developmental disorders due to its role in gene expression regulation. Abnormal RbBP5 activity can disrupt proper H3K4 methylation which may lead to either the inappropriate activation or silencing of genes implicating it in oncogenic processes. In cancer progression interactions between RbBP5 and other components like the MLL protein (mixed-lineage leukemia) become significant as mutations or dysregulation within this complex contribute to the pathogenesis of leukemia and other malignancies.
