Product details:
Abcam's TCA-3 (CCL1) Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an
in vitro
enzyme-linked immunosorbent assay designed for the quantitative measurement of TCA-3 (CCL1) in mouse serum, plasma and cell culture supernatants.
This assay employs an antibody specific for mouse TCA-3 coated on a 96-well plate. Standards and samples are pipetted into the wells and TCA-3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse TCA-3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and colour develops in proportion to the amount of TCA-3 bound. The Stop Solution changes the colour from blue to yellow, and the intensity of the colour is measured at 450 nm.

TCA-3 also known as CCL1 is a chemokine protein with an approximate molecular weight of 10 kDa. It belongs to the CC chemokine family and functions as a signaling molecule in immune responses. TCA-3 exhibits high expression in activated T lymphocytes macrophages and dendritic cells. This protein plays a role in the regulation of immune cell trafficking by interacting with specific G-protein-coupled receptors on the surfaces of target cells influencing cell movement towards the sites of inflammation or injury.
Biological function summary
TCA-3 impacts the immune system by contributing to cytokine receptor-mediated signaling pathways. It is involved in promoting T cell activation and differentiation facilitating immune surveillance and response. TCA-3 operates not as part of a complex but mainly influences cell behavior through its receptor CCR8. This interaction modulates key functions such as cell migration and immune response regulation.
Pathways
TCA-3 participates significantly in the chemokine signaling pathway and the innate immune response. It is vital in mediating inflammatory responses allowing TCA-3 to direct the movement of cells to inflammation sites. TCA-3 has functional links with proteins like CCR8 its receptor enabling T cells to respond selectively to TCA-3 signals. These associations highlight its role in shaping the body's ability to respond to immune challenges.
The dysregulation of TCA-3 has implications in conditions like rheumatoid arthritis and certain allergic reactions. Elevated levels of TCA-3 can lead to excessive recruitment of inflammatory cells exacerbating disease symptoms. Through its receptor CCR8 TCA-3 may influence disease progression and severity by altering immune cell behavior. Understanding TCA-3's role and connections in these disorders can inform therapeutic strategies targeting immune and inflammatory processes.
