TRAP80 also known as Mediator complex subunit 13 (MED13) or Thyroid Hormone Receptor-Associated Protein 80 kDa (THRAP80) plays an essential role in transcription regulation. It is a part of the Mediator complex and has a molecular mass of approximately 80 kDa. TRAP80 functions as a bridge connecting transcriptional activators bound at enhancer regions to the RNA polymerase II machinery enabling transcription initiation. Expression of TRAP80 occurs in various human tissues including heart brain and liver where it mediates gene expression responsive to hormonal signals.
Biological function summary
TRAP80 is involved in regulating gene expression by modifying the chromatin structure which impacts transcription by making specific genes accessible or inaccessible to RNA polymerase II. As a part of the Mediator complex TRAP80 participates in facilitating interactions between enhancers and promoters influencing transcription rates. Through these actions TRAP80 impacts processes like cell growth differentiation and response to external signals and it maintains proper cellular function by regulating gene networks.
Pathways
TRAP80 notably engages in the Wnt signaling pathway and thyroid hormone signaling pathway where it helps modulate transcriptional responses by activating or repressing target genes. In the Wnt pathway TRAP80 works with beta-catenin an important mediator to regulate target gene expression linked with cellular proliferation and differentiation.
