Abcam's Plasminogen Rat ELISA Kit is a two-site enzyme-linked immunosorbent assay (ELISA) for the quantitative measurement of Plasminogen levels in rat serum and plasma.
In this assay the Plasminogen present in samples reacts with the anti-Plasminogen antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-Plasminogen antibodies conjugated with horseradish peroxidase (HRP), are added. These enzyme-labeled antibodies form complexes with the previously bound Plasminogen. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of Plasminogen in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of Plasminogen in the test sample. The quantity of Plasminogen in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

Plasminogen also known as PLG is an important protein with a molecular weight of approximately 92 kDa. Plasminogen is a zymogen which means it is an inactive precursor that requires activation to function. It is produced primarily in the liver and circulates in blood plasma. Its expression occurs mainly in the liver but you can detect it in various tissues including the kidney. Plasminogen binds to tissue surfaces and undergoes transformation into plasmin an active enzyme that plays significant roles in many biological processes.
