The Epstein-Barr nuclear antigen 3B often referred to as EBNA 3B is a protein expressed by the Epstein-Barr virus (EBV). This protein is encoded within the EBV genome and shows an approximate molecular weight of 115 kDa. EBNA 3B is mainly expressed in EBV-infected B cells and plays a role in modulating the viral latent phase. It acts as a transcriptional regulator influencing the expression of both viral and host genes. Researchers also know the target by its role in latency and cellular transformation distinguishing it from other EBNA proteins.
Biological function summary
EBNA 3B contributes to the persistence of the virus in the host by modulating immune responses. It belongs to the EBNA 3 family which includes EBNA 3A and EBNA 3C forming a complex important for proper function. EBNA 3B modifies the host's cell cycle control and apoptosis aiding to maintain the viral latency essential for EBV survival and replication. This manipulation of the host cell environment ensures EBV evasion from immune system detection solidifying its role in persistent infection.
Pathways
EBNA 3B significantly influences the cell cycle and apoptosis pathways. It interacts with other EBNA proteins such as EBNA 2 and EBNA-LP in the Notch signaling pathway which plays an essential role in cell fate determination and immune response regulation. Through the modulation of these pathways EBNA 3B helps facilitate the long-term survival and proliferation of infected B cells. It supports a balance between viral latency and the risk of oncogenic transformation indicative of its strategic role in viral pathobiology.
EBNA 3B shows associations with lymphoproliferative disorders and certain types of cancers such as Burkitt's lymphoma. The protein establishes connections with other EBNA components like EBNA 1 and EBNA 2 which contribute to the oncogenic potential of EBV. These interactions can result in the deregulation of cellular growth and differentiation pathways promoting abnormal cell proliferation. Studies aim to better understand EBNA 3B's involvement in these conditions as targeting EBNA proteins holds therapeutic potential in EBV-related malignancies.
