EBNA 3C or Epstein-Barr virus nuclear antigen 3C is a significant protein encoded by the Epstein-Barr virus. Weighing approximately 100 kDa EBNA 3C operates within infected B cells. It plays an important role in transcriptional regulation interacting with various cellular proteins to modify gene expression. The protein recruits histone modification enzymes influencing chromatin structure and affecting transcriptional activity. EBNA 3C is often associated with the modulation of host cell cycle progression and can influence cell proliferation.
Biological function summary
The EBNA 3C protein contributes to the transformation of infected B cells into immortalized cells a hallmark of latent Epstein-Barr virus infection. This protein is part of an EBNAs family complex that includes EBNA 1 EBNA 2 and EBNA LP working together to alter host cell functions to the virus’s advantage. EBNA 3C has a role in the proteasome-dependent degradation of certain cellular proteins impacting cell cycle checkpoints and DNA repair mechanisms.
Pathways
EBNA 3C interacts with pathways that regulate cell cycle and apoptosis. The protein interrupts the retinoblastoma (Rb) protein pathway by degrading Rb-associated proteins allowing cell cycle progression independent of regular controls. Additionally the NF-kB signaling pathway where EBNA 3C acts enhances survival and proliferation signals in infected cells. The protein partnerships in these pathways include p53 and cyclin D1 which receive modulation of their functions by EBNA 3C to favor viral persistence.
EBNA 3C has associations with various Epstein-Barr virus-related cancers such as Burkitt's lymphoma and nasopharyngeal carcinoma. In these diseases EBNA 3C modulates oncogenic pathways supporting uncontrolled cell growth. The protein's interaction with p27 which regulates cell cycle progression further connects EBNA 3C to oncogenic processes.
