The protein arginine methyltransferase 8 commonly referred to as PRMT8 catalyzes the transfer of methyl groups to arginine residues on target proteins. It is part of the PRMT family and possesses a molecular mass of approximately 42 kDa. PRMT8 shares sequence similarity with PRMT1 but shows unique tissue-specific expression. Notably it is mainly found in the brain where it localizes to the plasma membrane suggesting specialized functions in neuronal cells.
Biological function summary
PRMT8 contributes to modulating signal transduction pathways in neurons. It plays a role in the post-translational modification of proteins which affects their interaction and stability. Though not known to form any stable large complexes PRMT8's methyltransferase activity modifies substrate proteins influencing their cellular functions. In neurons this activity may regulate synaptic plasticity or neural communication through methylation of proteins involved in signal transduction.
Pathways
Arginine methylation by PRMT8 influences several key signaling pathways including the phosphoinositide and protein kinase pathways. PRMT8's activity affects proteins such as Src family kinases which are important in transmitting signals for cell growth and differentiation. Interaction with these pathways suggests that PRMT8 serves as a modulator of complex cellular signaling mechanisms balancing neuronal response and adaptation.
Abnormalities in PRMT8 activity have associations with neurodegenerative diseases and brain-related disorders including Alzheimer's disease. Dysregulation of PRMT8 may affect amyloid precursor protein processing implicating its role along with proteins such as tau in disease pathology. Understanding PRMT8's involvement in these conditions could inform therapeutic strategies that target arginine methylation to ameliorate or prevent disease progression.
