CHAMP also known as Chromatin Associated Module Protein serves an important role in chromatin remodeling. Its molecular mass is approximately 75 kDa. This protein localizes mainly in the nucleus where it assists in altering chromatin structure to regulate DNA access. CHAMP facilitates the binding of transcription factors by repositioning nucleosomes which is essential for gene expression and DNA repair processes.
Biological function summary
CHAMP plays a role in the regulation of gene expression and impacts cellular differentiation and proliferation. It acts as part of a larger chromatin remodeling complex that includes several other proteins enabling the coordination of multiple functions necessary for cellular homeostasis. Its activity ensures that specific genes are either activated or repressed as the cell requires contributing to the diversity of cellular functions.
Pathways
CHAMP contributes to key signaling cascades such as the Wnt signaling and Notch signaling pathways. In the Wnt pathway its modification of chromatin accessibility allows transcription factors like TCF/LEF to bind DNA and activate transcription. The protein shows interactions with core components like β-catenin in Wnt signaling and in the Notch pathway it influences cellular fate decisions by impacting expression of target genes critical for development.
Improper function of CHAMP has connections to cancer and neurological disorders. Altered activity can lead to dysregulation of gene expression promoting tumorigenesis where it often associates with increased levels of β-catenin an important protein in cancer development. Similarly in neurological disorders its failure in chromatin modification affects the expression of genes essential for neural function sometimes involving interactions with proteins like NICD in Notch-related processes.
