ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of mouse Angiopoietin-2 in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA).
An Angiopoietin-2 specific rat monoclonal antibody has been precoated onto 96-well plates. Standards and test samples are added to the wells and incubated. A biotinylated detection polyclonal antibody from goat, specific for Angiopoietin-2 is then added followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. TMB is then used to visualize the HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the mouse Angiopoietin-2 amount of sample captured in plate.

Angiopoietin-2 also known as ANG2 or ANGPT2 is a glycoprotein with a molecular mass of approximately 70 kDa. It plays an important role in angiogenesis facilitating blood vessel remodeling and maturation. The protein is mainly expressed in endothelial cells where it functions by binding to the Tie2 receptor partially disrupting the Angiopoietin-Tie2 signaling axis. This disruption modulates vascular stability and permeability. The lyophilized form of the ANG-2 protein is often used in research for functional assays like ANG-2 ELISA to quantify its levels in various biological samples.

Human angiogenesis processes are significantly affected by Angiopoietin-2 which serves as an antagonist to Angiopoietin-1 in regulating vascular dynamics.
