Rad51C also known as RAD51 homolog C is a protein involved in the repair of DNA double-strand breaks through homologous recombination. It plays a mechanical role by forming nucleoprotein filaments essential for DNA pairing and strand exchange processes. The mass of Rad51C is approximately 42 kDa. This protein expresses widely in various tissues including ovaries and testes indicating its importance in genomic stability and cell division.
Biological function summary
Rad51C contributes to maintaining genomic integrity by participating in the formation of the Rad51 paralog complex. This complex is essential in the repair of DNA damage and supports replication fork stability. Rad51C collaborates with other paralogs such as Rad51B Rad51D and XRCC2 forming the BCDX2 complex which promotes homologous recombination and protects against genotoxic stress. This involvement highlights its critical role in cellular defense mechanisms against DNA damage.
Pathways
The protein participates actively in the homologous recombination repair pathway and the DNA damage response pathway. Within these pathways Rad51C interacts with pivotal genes such as BRCA1 BRCA2 and PALB2 facilitating repair of DNA damages and coordinating cellular response to replication stress. The interactions with these proteins form a network that orchestrates precise DNA repair processes ensuring cellular survival and minimization of mutations.
Rad51C connections include hereditary breast and ovarian cancer syndrome and Fanconi anemia-like phenotypes. Mutations or deficiencies in Rad51C can lead to compromised DNA repair raising the risk of these malignancies. In the context of cancer Rad51C operates alongside BRCA1 and BRCA2 where dysfunctions may result in elevated susceptibility to these conditions. Recognizing these interactions aids in understanding cancer pathogenesis and potential therapeutic targets.
