The HuB HuC and HuD proteins often referred to collectively as Hu antigens are neuronal RNA-binding proteins. They belong to the ELAVL (Embryonic Lethal Abnormal Vision in Drosophila) protein family. These proteins have masses typically ranging from around 35-42 kDa. They are expressed chiefly in the nervous system where they play critical roles in the regulation of mRNA. HuB (also known as ELAVL2) HuC (ELAVL3) and HuD (ELAVL4) support neuronal differentiation and maintenance.
Biological function summary
The Hu antigens stabilize and modulate the translation of target mRNAs impacting processes such as neuronal growth and synaptic plasticity. These proteins do not exist in isolation; they are integrated into RNP (ribonucleoprotein) complexes in neurons. HuC and HuD in particular serve as important neuronal markers often used in research to identify neuronal lineage and differentiation. Hu proteins influence gene expression by binding to AU-rich elements located in the 3' untranslated region of mRNAs.
Pathways
HuB HuC and HuD play a significant role in the post-transcriptional regulation pathway influencing the stability and translation of mRNAs involved in neuronal function. This is important for learning and memory processes where proteins like FMRP are also involved. HuD for instance is linked with pathways that regulate mRNA turnover and translation initiation affecting how neural cells respond to various stimuli.

Abnormal interactions between HuD and tau protein which is implicated in Alzheimer's suggest pathways where Hu proteins contribute to disease progression.
