Zinc Alpha 2 Glycoprotein (ZAG) also called ZAG or AZGP1 is a soluble protein with a molecular mass of approximately 41 kDa. It functions as an adipokine and is mostly found in the liver breast and prostate tissues as well as in bodily fluids like plasma and saliva. ZAG belongs to the major histocompatibility complex class I family and it shares structural similarities with antigen-presenting proteins.
Biological function summary
Zinc Alpha 2 Glycoprotein assists in lipid metabolism. It promotes the breakdown of triglycerides in adipose tissue by stimulating the beta-adrenergic pathway. ZAG functions independently and does not typically form complexes with other proteins during this process. Its presence in adipose tissue suggests a role in energy homeostasis and body weight regulation.
Pathways
Zinc Alpha 2 Glycoprotein participates in the metabolic pathway for lipid degradation. The protein interacts with beta-3 adrenergic receptors to trigger lipolysis which involves breaking down fat cells into free fatty acids and glycerol. Its action in this pathway links ZAG with other proteins such as perilipin which protects lipid droplets from breakdown.
Zinc Alpha 2 Glycoprotein has connections with metabolic-related conditions notably obesity and cancer cachexia. In obesity lower levels of ZAG are often observed potentially linking it to impaired fat metabolism. In cancer cachexia elevated ZAG levels are frequently detected suggesting a role in the loss of fat and muscle tissue. ZAG's interaction with certain cytokines may enhance cachexia progression by affecting protein degradation pathways.
