Kallikrein 4 also known by the alternate name KLK4 is a serine protease enzyme with a molecular mass of approximately 30 kDa. This enzyme belongs to the kallikrein family which is a subset of the larger protease family. KLK4 is mainly expressed in the prostate teeth and skin. Its primary mechanical function involves cleaving peptide bonds in proteins contributing to protein degradation and activation of other proteins.
Biological function summary
KLK4 plays a significant role in the remodeling of extracellular matrix components. It is particularly involved in dentin matrix degradation during tooth development where it contributes to enamel formation and maturation. Although not part of a larger protein complex KLK4 works in conjunction with enzymes like KLK1 and KLK3 to regulate various biological processes through proteolytic action.
Pathways
The activity of KLK4 integrates into the kallikrein-kinin system and the broader ECM degradation pathway. Within the kallikrein-kinin system KLK4 functions alongside proteins like KLK1 to influence vasodilation and inflammation. In the ECM degradation pathway KLK4 contributes to matrix remodeling by breaking down structural proteins facilitating physiological processes like tissue repair and cancer metastasis.
KLK4 has connections with prostate cancer and amelogenesis imperfecta. In prostate cancer aberrant expression of KLK4 might contribute to tumor progression associating with prostate-specific antigen (PSA) also known as KLK3. In amelogenesis imperfecta mutations in the KLK4 gene can impair enamel formation emphasizing its role in tooth integrity and formation.
