Rotavirus also called RVA is a non-enveloped virus that primarily infects the gastrointestinal tract. It has a double-stranded RNA genome encoding six structural proteins and six non-structural proteins. The virus particles have a triple-layered capsid which is 70-75 nanometers in diameter. Rotavirus is expressed in the small intestine of humans and animals where it attaches to and enters mature enterocytes. The virus disrupts these cells’ function and leads to fluid loss and diarrhea.
Biological function summary
Rotavirus significantly impacts intestinal absorption and secretion balance. The virus produces non-structural protein 4 (NSP4) which functions as an enterotoxin. NSP4 alters intracellular calcium homeostasis and induces secretion resulting in diarrhea. Rotavirus is not part of a complex; its independent proteins perform roles that contribute to its pathogenicity and replication.
Pathways
Rotavirus infection affects host signaling pathways like NF-κB and interferon (IFN) responses. Disruption of these pathways helps the virus to circumvent immune elimination and propagate in the host. NSP4 interacts with host extracellular signal-regulated kinases (ERK) influencing cellular processes that give the virus a replication advantage. These pathways signal cellular responses that affect inflammation and cell apoptosis.
Rotavirus is mainly implicated in acute gastroenteritis especially in children under five. This condition can lead to severe dehydration and in extreme cases can be life-threatening without adequate treatment. There is a relation between Rotavirus and the protein β-defensin which the virus counteracts to evade innate immune defenses. Vaccination has proven effective in reducing disease incidence and associated healthcare burdens worldwide.
