TAGAP also known as T-cell activation RhoGTPase activating protein acts as a Rho GTPase-activating protein involved in the regulation of the actin cytoskeleton. It weighs approximately 70 kDa. This protein is particularly expressed in immune cells including T-cells B-cells and dendritic cells. TAGAP plays an important role in the modulation of signaling pathways essential for immune cell activation and function.
Biological function summary
TAGAP contributes to the regulation of immune responses. It influences the rearrangement of the actin cytoskeleton affecting cell movement and activation. Though not a part of a large protein complex TAGAP's ability to manage cytoskeletal dynamics is critical in immune cells such as T-cells enabling proper response to antigens.
Pathways
TAGAP is significant in the T-cell receptor signaling and actin cytoskeleton regulation pathways. These pathways facilitate immune cell activation and migration key processes in immune response. TAGAP mediates interactions with proteins like Rac1 involved in cytoskeletal organization and Vav1 a guanine nucleotide exchange factor that activates pathways necessary for the immune response.
TAGAP's function links to autoimmune diseases like multiple sclerosis and rheumatoid arthritis. Altered TAGAP expression or function can influence disease susceptibility making it a target for research. Through autoimmune-related pathways TAGAP interacts with proteins such as IL-2 and STAT4 which are implicated in these disorders highlighting its potential role in influencing immune system balance and disease progression.
