ANKLE1 also known as Ankyrin Repeat and LEM Domain containing 1 or LEM3 is a protein that functions as a DNA endonuclease. This protein has a molecular mass of approximately 63 kDa. It is expressed in various tissues with presence reported in the testis ovaries placenta and to a lesser extent in other tissues. ANKLE1 contains ankyrin repeat domains which often mediate protein-protein interactions. Through its mechanical action it is involved in DNA damage response.
Biological function summary
ANKLE1 acts in the maintenance of genomic stability. It plays a role in resolving DNA structures during replication and repair processes. Evidence suggests ANKLE1 does not function as part of a larger protein complex. Instead it performs its endonuclease activity individually to protect cells from DNA damage. This activity makes it essential for proper cell cycle progression and preventing genomic abnormalities.
Pathways
Functional aspects of ANKLE1 integrate into the DNA damage repair pathway specifically the homologous recombination repair pathway. Its endonuclease action helps in processing DNA intermediates that arise during repair. ANKLE1 works in conjunction with other proteins such as RAD51 which facilitates the strand invasion step of homologous recombination. This collaboration helps ensure genomic integrity is preserved during cell division.
Aberrations in ANKLE1 function or expression have associations with certain cancers such as breast cancer. Its role in safeguarding genomic integrity hints at its protective nature against tumorigenesis. Connections between ANKLE1 and proteins like BRCA1 a well-known key player in DNA repair processes illustrate its potential importance in maintaining genomic stability. Studies continue to explore its precise implications in cancer biology and any potential as a therapeutic target.
