PVL LukS also known as Panton-Valentine leukocidin S component is a part of a bacterial toxin produced by certain strains of Staphylococcus aureus. It has a molecular mass of approximately 32.9 kDa. This protein is expressed mainly in pathogenic strains of Staphylococcus aureus making it an important factor for assessing bacterial virulence. PVL LukS works closely with another component LukF and together they create pores in the membranes of host immune cells especially leukocytes causing cell lysis.
Biological function summary
PVL LukS forms a binary toxin complex by pairing with the LukF component. This complexly binds to specific receptors on the surface of human immune cells. Once bound the two components oligomerize to form a transmembrane pore. The formation of these pores disrupts cell membrane integrity leading to cell death and tissue damage which contributes significantly to the pathogenesis of the infection caused by Staphylococcus aureus.

PVL LukS participates mainly in immune cell evasion strategies of the bacteria. It plays a role in pathways that disrupt host immune function by targeting and destroying leukocytes which are significant players in the body's defense mechanism. Its interaction with the protein LukF is integral as their combined action enables the formation of the apoptogenic complex on host cell surfaces. This pathway is a part of the virulence strategy employed by Staphylococcus aureus to induce local infection and evade the immune response.
The PVL LukS protein is associated with severe skin and soft tissue infections as well as necrotizing pneumonia. These conditions arise due to the lytic action of the PVL toxin on immune cells weakening the host's defense and allowing the bacteria to spread. The target is connected to other virulence factors of Staphylococcus aureus such as protein A which interferes with host immune recognition thereby compounding the severity of infections associated with PVL.
