The replication factor C subunit 4 also known as RFC4 is an essential protein in DNA replication possessing a molecular weight of approximately 37 kDa. RFC4 forms part of the larger replication factor C (RFC) complex which functions as a clamp loader for the proliferating cell nuclear antigen (PCNA). This activity facilitates the attachment of PCNA to the DNA promoting DNA polymerase processivity during replication. RFC4 is widely expressed across various tissues with higher levels noted in proliferating cells.
Biological function summary
RFC4 plays a critical role in maintaining genomic stability and facilitating proper cell cycle progression. As a component of the RFC complex it cooperates with additional subunits to engage with DNA replication machinery effectively. This complex recognizes the primer-template junction and recruits PCNA to assist DNA polymerase in duplicating the genome accurately. RFC4's role does not end with replication as the RFC complex also partakes in DNA repair mechanisms needed for correcting replication errors.
Pathways
RFC4 is actively involved in the DNA replication pathway and the DNA damage response pathway. Within these pathways it interacts with proteins like DNA polymerase delta and epsilon ensuring their efficient function during DNA synthesis and repair. RFC4 collaborates closely with PCNA in these pathways to maintain fidelity in replication and in responding to DNA damage highlighting its functional importance in genetic information preservation.
RFC4 holds significance in the context of cancer and genomic instability disorders. Dysregulation or mutations in RFC4 can lead to defective DNA replication and repair contributing to uncontrolled cell proliferation seen in cancer. Its interaction with other proteins such as PCNA and DNA polymerases can become aberrant further exacerbating disease progression. Research into RFC4 offers potential insights into therapeutic strategies that target replication and repair pathways for disease intervention.
