TRIM23 also known as ARD1 is a protein with an approximate mass of 77 kDa. It belongs to the tripartite motif (TRIM) family which is characterized by the presence of a RING finger domain a B-box domain and a coiled-coil region. TRIM23/ARD1 is expressed in various tissues including the heart and skeletal muscle. Its role as an E3 ubiquitin ligase is vital as it facilitates the ubiquitination and proteasomal degradation of specific substrates contributing to protein homeostasis within cells.
Biological function summary
TRIM23/ARD1 plays a role in immune and inflammatory responses. It participates in the maturation of autophagosomes a process that is essential for cellular repair and stress responses. Rather than functioning independently TRIM23/ARD1 associates with complex cellular systems including the endosomal-lysosomal pathway influencing the degradation and recycling of cellular components.
Pathways
TRIM23/ARD1's activity is integral to the autophagy and NF-kB signaling pathways. In the autophagy pathway TRIM23/ARD1 acts alongside proteins like ULK1 regulating autophagosomal formation and turnover. Within the NF-kB signaling pathway TRIM23/ARD1 modulates transcription factors that respond to inflammation and stress. Through these pathways TRIM23/ARD1 influences cellular responses to environmental stimuli ensuring proper cellular function.
TRIM23/ARD1's malfunctions have associations with cancer and neurodegenerative diseases. Its role in cancer involves the regulation of cell proliferation and apoptosis; disruptions can contribute to tumor growth. In neurodegenerative diseases TRIM23/ARD1 interacts with proteins such as tau impacting protein aggregation and clearance which are critical in diseases like Alzheimer's. Understanding TRIM23/ARD1's interactions in these conditions could provide insights into potential therapeutic targets.
