The SHANK3 protein also known as ProSAP2 is a scaffold protein that plays an important role at the postsynaptic density of excitatory synapses. It has a mass of around 240 kDa. SHANK3 is expressed mainly in the brain but also present in heart spleen and kidneys. It interacts with various proteins through its multiple domains contributing to the structural integrity and signaling functions of synapses.

SHANK3 connects the cytoskeleton to the membrane proteins maintaining synaptic structure and function. As a part of the postsynaptic density complex SHANK3 interacts with proteins like Homer and NMDA receptors to organize the signaling platforms.
