Abcam's TNFSF4 Mouse ELISA Kit (ab193729) is an
in vitro
enzyme-linked immunosorbent assay for the quantitative measurement of mouse TNFSF4 in serum, plasma and cell culture supernatants.
This assay employs an antibody specific for mouse TNFSF4 coated on a 96-well plate. Standards and samples are pipetted into the wells and the immobilized antibody captures TNFSF4 present in the samples. The wells are washed and biotinylated anti-mouse TNFSF4 antibody is added. After washing away any unbound biotinylated antibody, an HRP-conjugated streptavidin is pipetted to the wells. After incubation, the wells are again washed, followed by the addition of a TMB substrate solution to the wells. Color will develop in proportion to the amount of TNFSF4 bound in each well. Addition of the Stop Solution will change the color from blue to yellow, and the intensity of the color is measured at 450 nm.

OX40L also known as TNFSF4 is a member of the tumor necrosis factor (TNF) ligand family. It is a type II transmembrane protein with a molecular weight of about 32 kDa. OX40L is expressed mainly on activated antigen-presenting cells such as B cells dendritic cells and macrophages. The protein interacts specifically with its receptor OX40 (CD134) which is found on activated T cells mainly CD4+ T cells. These interactions are important for the regulation of immune responses.

OX40L acts as a costimulatory signal for T cells. It does not work alone; it functions as part of the immune complex involved in enhancing T cell proliferation cytokine production and survival.
