The EZH2 + EED + SUZ12 complex also called the Polycomb Repressive Complex 2 (PRC2) plays a significant role in gene silencing through methylation of histone H3 at lysine 27 (H3K27me3). EZH2 acts as the catalytic subunit while EED and SUZ12 stabilize and enhance its catalytic activity. The mass of EZH2 is approximately 83 kDa. This complex is usually expressed in the nucleus where it performs its function in epigenetic regulation.
Biological function summary
PRC2 is essential for maintaining the silenced state of target genes. It acts as a part of the Polycomb-group (PcG) proteins which are important for transcriptional repression. Through their activity these complexes preserve the repression of developmental genes and prevent abnormal activation during cell differentiation and proliferation. PRC2 often teams up with other chromatin modifiers and DNA-binding proteins to accurately target specific genomic loci.
