JIP3 also known as c-jun-amino-terminal kinase-interacting protein 3 or MAPK8IP3 is a scaffold protein with a molecular mass of approximately 150 kDa. It mediates intracellular signaling pathways and facilitates interactions between kinases and their substrates. JIP3 predominantly localizes in the cytoplasm and expresses highly in the brain especially in neurons. This protein plays a role in axonal transport and cytoskeletal organization.
Biological function summary
JIP3 influences neuronal signaling and development processes. It acts as a scaffold in kinase cascades particularly within the JNK signaling pathway forming complexes with JNK and other MAP kinases. The protein ensures proper localization and docking of kinases promoting efficient signal transduction. JIP3 links signaling components thereby influencing processes like neuronal migration and polarization.
Pathways
JIP3 serves as a significant component in the JNK and p38 MAPK pathways. These pathways are important for regulating cellular responses to stress apoptosis and immune responses. Within these pathways JIP3 interacts with JNK and other associated proteins like MAPK8 facilitating proper signaling flow. JIP3's function as a molecular scaffold coordinates cellular events through these signal transduction pathways.
JIP3 connects to neurodegenerative diseases such as Alzheimer’s and Amyotrophic Lateral Sclerosis (ALS). Dysregulation in the JNK signaling pathway often links to these conditions. Additionally disturbed axonal transport involving JIP3 can contribute to such neurodegenerative disorders. The relationship of JIP3 with other proteins like tau in Alzheimer’s further highlights the involvement of JIP3 in disease pathogenesis.
