{EGR4} also known as Early Growth Response 4 is a transcription factor involved in the regulation of gene expression. It belongs to the EGR family of C2H2-type zinc-finger proteins. EGR4 has a mass of approximately 59 kDa and expresses in a variety of tissues including the brain testis and other nervous tissues. The protein serves as an important regulator in neuronal differentiation and development playing a role in synaptic plasticity and cognitive functions.
Biological function summary
The Early Growth Response 4 protein modulates the transcription of genes involved in cellular growth and apoptosis. It does not work in large complexes but often collaborates with other transcription factors and cofactors. This interaction helps control the expression of genes necessary for cell proliferation differentiation and stress response. EGR4’s activity significantly affects the balance between cell survival and death impacting cellular homeostasis and adaptation to external signals.
Pathways
EGR4 interacts with multiple signal transduction pathways. Notably it engages in the MAPK/ERK pathway which plays a critical role in transmitting growth signals from the cell surface to the nucleus. Moreover EGR4 influences the PI3K/AKT pathway related to cell survival and metabolism. These pathways connect EGR4 with other proteins such as c-JUN and CREB important for gene regulation and cellular response mechanisms.
The dysregulation of EGR4 has been associated with certain neurological disorders and cancers. In particular aberrant expression of EGR4 links to Alzheimer's disease where it might contribute to neuronal damage and impaired cognitive functions. Additionally EGR4 involvement appears in the progression of testicular tumors indicating its role in cell proliferation and malignancy. EGR4's interaction with p53 a prominent tumor suppressor protein could exacerbate or mitigate these disease processes by influencing apoptosis or cell cycle regulation.
