MAKAP also known as muscle A-kinase anchoring protein plays an important role in cellular signaling by anchoring protein kinase A (PKA) and other signaling proteins. It has a molecular weight of approximately 250 kDa. mAKAP is primarily expressed in striated muscle tissues including cardiac and skeletal muscles. Its localization within the cell is important for its function as it coordinates the activity of various signaling molecules in response to external and internal stimuli.
Biological function summary
MAKAP facilitates the integration of cyclic adenosine monophosphate (cAMP)-dependent signaling pathways by anchoring PKA and phosphodiesterases (PDEs) within signaling complexes. It organizes these complexes at the perinuclear region and other cellular sites ensuring precise signaling. By supporting the assembly of these molecular components mAKAP regulates processes such as contractility and hypertrophy in cardiac muscle.
Pathways
MAKAP acts as a scaffold that coordinates the cAMP and calcium signaling pathways which are critical in cardiac muscle function. It interacts with PKA and PDEs to finely tune the signaling cascades that control heartbeats and muscle contraction. Furthermore mAKAP has a functional relationship with the small GTPase Ras integrating signaling inputs that modulate cell cycle and growth responses.
MAKAP has been implicated in cardiac diseases particularly in heart failure and cardiac hypertrophy.
