ENO4 an abbreviation for Enolase 4 is an enzyme that participates in the glycolytic pathway. This protein also known as testis-specific enolase has a mass of approximately 50 kDa. ENO4 expresses mainly in the testis although low levels appear in other tissues. Its functional role involves catalyzing the reversible conversion of 2-phosphoglycerate to phosphoenolpyruvate an essential step in glycolysis which plays an important role in energy production.
Biological function summary
ENO4 is important for energy metabolism within cells particularly in spermatogenesis. During this process ENO4 contributes to the development and maturation of sperm cells by ensuring efficient energy supply. While not a primary component of any large protein complexes ENO4 interacts with other glycolytic enzymes and requires co-factors like magnesium ions for its enzymatic activity. Its specificity and expression pattern make it distinctive among the enolase family members.
Pathways
ENO4 plays a significant role in the glycolytic pathway one of the central energy-producing processes in cells. This protein connects functionally with other glycolytic enzymes like phosphogluconate dehydrogenase and pyruvate kinase. These interactions ensure the sequential breakdown of glucose to generate ATP which is important for cell proliferation and energy maintenance especially in the context of spermatogenesis.
ENO4 has associations with male infertility and certain metabolic disorders. Due to its specific expression in the testis and role in sperm development alterations in ENO4 expression or function can lead to abnormalities in sperm production affecting male fertility. Moreover disruptions in glycolytic enzymes including ENO4 may contribute to metabolic syndromes although studies often focus on more common glycolytic enzymes. Understanding its interaction with related glycolytic proteins such as phosphogluconate dehydrogenase helps in clarifying its involvement in these conditions.
