CGNL1 also known as Paracingulin is a protein that plays a significant mechanical role in cellular junction assembly and maintenance. It is approximately 155 kDa in mass and functions as a cytoplasmic protein. CGNL1 localizes at tight and adherens junctions and is widely expressed in epithelial tissues. This target is involved in the stabilization of cell-cell interactions contributing to the structural integrity of cellular sheets important for epithelial function.
Biological function summary
CGNL1 contributes to the control of cell junction dynamics and is part of the cingulin family complex. CGNL1 is important in linking cytoskeletal components to junctional complexes allowing cells to maintain their polarity and adhesion. It interacts with various proteins such as ZO-1 and JAM-A to coordinate the assembly and disassembly of junctional structures. These interactions facilitate the proper functioning of tight junctions which are essential for the paracellular barrier and selective permeability.
Pathways
CGNL1 participates in the Rho GTPase signaling and Wnt signaling pathways. In the Rho GTPase pathway CGNL1 interacts with the small GTPases including RhoA and Rac1 to regulate cytoskeletal organization and junction formation. The Wnt signaling pathway connects CGNL1 to the regulation of cell proliferation and differentiation where it collaborates with proteins like beta-catenin to ensure proper signal transduction. This emphasizes its role in maintaining cellular structural integrity and communication within tissues.
CGNL1 has been linked to cancer and inflammatory bowel disease (IBD). Alterations in CGNL1 expression or function can disrupt junctional complexes leading to increased permeability and a compromised epithelial barrier which are observed in IBD. In cancer abnormal CGNL1 levels can influence cell migration and invasion contributing to tumor progression. CGNL1 interacts with proteins such as occludin and claudins connecting it to these disorders and emphasizing its impact on cellular junction integrity.
