Polycystin 2 also known as PKD2 or TRPP2 acts as a non-selective cation channel involved in calcium signaling with a molecular mass of approximately 110 kDa. This channel is part of the transient receptor potential (TRP) superfamily and is expressed in multiple tissues including the kidney heart liver and pancreas. As an integral membrane protein Polycystin 2 contributes to ion homeostasis and cellular signaling.
Biological function summary
The protein controls calcium release from intracellular stores and regulates various cellular processes. Polycystin 2 often forms a functional complex with Polycystin 1 affecting its location and activity. This dimerization is significant for mechanosensation and flow sensing in renal epithelial cells. This protein complex participates in maintaining tubular structure and function which is essential for proper kidney operation.
Pathways
Polycystin 2 plays roles in the Wnt signaling and mTOR pathways which are important for cell proliferation and differentiation. In the Wnt signaling pathway Polycystin 2 modulates intracellular calcium levels influencing pathway-related responses. It interacts with other proteins such as Polycystin 1 and mTOR mediating growth factor signals and cellular metabolism. These interactions help coordinate response to mechanical stress and regulate cell cycle progression.
Defects in Polycystin 2 relate to autosomal dominant polycystic kidney disease (ADPKD) and cardiovascular abnormalities. ADPKD arises from mutations in the PKD2 gene leading to cyst formation and kidney dysfunction. Polycystin 2 interacting with Polycystin 1 serves an important function in preventing these cystic abnormalities. Moreover irregularities in calcium signaling caused by defective Polycystin 2 can result in altered heart rhythm and other cardiovascular issues.
