KBTB6 also known as KBTBD6 is a protein that functions as an E3 ubiquitin ligase specifically targeting proteins for ubiquitination and subsequent proteasomal degradation. The protein has a molecular mass of approximately 64 kDa. KBTB6 is expressed in various tissues with noticeable levels in the liver and muscle tissues. It interacts with Cullin-RING ubiquitin ligase complexes facilitating the transfer of ubiquitin molecules to substrates which marks them for degradation.
Biological function summary
The E3 ligase activity of KBTB6 regulates protein homeostasis by degrading unnecessary or damaged proteins. It plays an essential role in several cellular processes including cell cycle regulation signal transduction and cellular differentiation. KBTB6 works as part of a larger complex with other proteins like cullins and RING box proteins to perform its function effectively. These complexes ensure the specificity of substrate identification and ubiquitination.
Pathways
KBTB6 influences various cellular pathways by regulating protein stability. It is involved in the Wnt signaling pathway and also links to the p53 pathway by modulating the levels of critical regulatory proteins within these pathways. In these roles KBTB6 interacts with proteins like beta-catenin in the Wnt pathway and Mdm2 in the p53 pathway which are important for the regulation of cellular proliferation and apoptosis.
Disruptions in KBTB6 function can relate to cancer and muscular disorders. Its role in regulating the degradation of proteins involved in cell cycle and apoptosis makes it significant in tumorigenesis where alterations can lead to uncontrolled cell growth. Also abnormalities in KBTB6 activity are associated with certain muscle diseases due to its expression in muscle tissues. In these conditions it may interact with other proteins such as p53 or beta-catenin which play roles in cell regulation and signal transduction processes.
