MafB also known as V-maf musculoaponeurotic fibrosarcoma oncogene homolog B acts as a transcription factor involved in the regulation of gene expression. The protein has a molecular weight of approximately 37 kDa. MafB localizes predominantly to the nucleus where it exerts its function. Expression of MafB occurs in various tissues with significant levels in hematopoietic cells kidney and pancreas reflecting its role in cell differentiation and organ development.
Biological function summary
MafB plays an important role in the regulation of monocyte and macrophage differentiation. It serves as an important regulator that controls genes required for the immune response and terminal differentiation of these cells. MafB often operates as a heterodimer pairing with other transcription factors like Fos and Jun to form complexes that specifically bind to Maf recognition elements in DNA. This binding allows it to orchestrate various transcriptional programs necessary for immune system function and organogenesis.
Pathways
MafB integrates into the larger network of macrophage differentiation and the insulin signaling pathway. It interacts closely with proteins like PU.1 in the macrophage differentiation pathway where it helps specify the lineage commitment of hematopoietic progenitor cells.
